A chia (Salvia hispanica L.) é uma planta herbácea cultivada anualmente, sendo considerada oleaginosa em virtude do alto teor de óleo, que corresponde de 30 a 40% da semente. As sementes de chia quando em presença de água possuem a capacidade de formar um gel transparente mucilaginoso devido à presença de moléculas hidrofílicas que se combinam com a água. A mucilagem de chia representa cerca de 6% da semente de chia e é composta principalmente por fibras dietéticas, as quais apresentam uma grande capacidade de retenção e absorção de água. Além disso, a mucilagem de chia apresenta baixo teor lipídico. Logo, a mucilagem de chia se mostra com um novo ingrediente, com potencial aplicabilidade em alimentos, atuando como emulsificante, estabilizador de espuma, agente de ligação e substituto de gordura. Diante disso, este trabalho teve como objetivo produzir mucilagem de chia (Salvia hispanica L.) para aplicação na substituição da gordura em produtos alimentícios, como pães, bolos de chocolate e maionese. Inicialmente, foi realizado um estudo da extração da mucilagem de chia, utilizando diferentes relações semente:água e tipos de secagem, seguida de sua caracterização. As mucilagens de chia liofilizada e seca a 50ºC foram aplicadas na substituição da gordura em pão e bolo de chocolate, já a mucilagem de chia liofilizada foi utilizada para a substituição do óleo e da gema de ovo em maionese. Os produtos desenvolvidos foram caracterizados quanto à composição proximal, suas características tecnológicas e resposta sensorial de aceitação. A substituição de até 75% da gordura nos pães pela adição da mucilagem de chia garantiu aumento no volume especifico e manteve as demais características tecnológicas, sendo que quando testadas sensorialmente, e nesse mesmo grau de substituição, os pães com adição da mucilagem de chia seca a 50ºC apresentaram maior aceitabilidade e maior intenção de compra. Os bolos de chocolate apresentaram resultados semelhantes ao dos pães, embora os melhores resultados foram obtidos com substituições inferiores a 50%. Sensorialmente, os bolos de chocolate com 75% da substituição da gordura por mucilagem de chia não apresentaram diferença significativa quanto ao tipo de mucilagem, entretanto a intenção de compra foi maior para mucilagem seca a 50ºC. Em relação às maioneses, a substituição do óleo por mucilagem de chia liofilizada apresentou aumento da estabilidade, em relação à amostra controle, e propriedades de textura semelhantes à maionese comercial. Já a adição de mucilagem de chia liofilizada pela substituição da gema de ovo apresentou resultados negativos, tanto para os parâmetros de textura, como na redução do teor lipídico. Sensorialmente, as maioneses com substituição da gema de ovo foram melhor aceitas do que as com substituição do óleo. Com este estudo, foi possível verificar que a mucilagem de chia apresenta propriedades que a possibilitam atuar como agente emulsificante em produtos alimentícios. Além disso, os produtos alimentícios desenvolvidos apresentaram-se mais saudáveis, com menor conteúdo de lipídios, garantindo características sensoriais e tecnológicas adequadas.; Chia (Salvia hispanica L.) is a herbaceous plant grown annually, and it is considered an oilseed because of the high oil content, which is 30-40% of the seed. The chia seed when in the presence of water have the capacity to form a transparent mucilaginous gel due to the presence of hydrophilic molecules which combine with water. The chia mucilage represents about 6% of chia seed and is composed mainly of dietary fibers which have a large holding and water absorption capacity. In addition, the chia mucilage has a low fat content. Therewith the chia mucilage shown with a new ingredient with potential applications in foods, acting as emulsifier, foam stabilizer, binding agent and fat replacer. Therefore, this study had as objective to produce mucilage of chia (Salvia hispanica L.) for use as fat replacer in food products such as breads, chocolate cakes and mayonnaise. Initially, it was conducted a study of the extraction of the chia mucilage, using different relations seed:water and two drying types, followed by their characterization. The chia mucilage in form of lyophilized and dried at 50°C were applied in the partial and total replacement of fat in bread , chocolate cake, and lyophilized chia mucilage was used for replacement of oil and egg yolk in mayonnaise. The developed products were characterized for proximate composition and its technological and physical characteristics. At the end, the sensory analysis of the products was performed. Replacement of up to 75% of the fat in breads by adding chia mucilage produced increase the specific volume and keeping the other technological characteristics, whereas when tested by sensory evaluation, products with the same degree of substitution, breads with addition of chia mucilage dried at 50ºC presented higher acceptability and purchase intent by the judges, compared with products substituted with lyophilized mucilage. The chocolate cakes showed similar results to breads, although the best results were obtained with level substitutions less than 50% of chia mucilage. In sensory evaluation, chocolate cakes with 75% of substitution of fat per chia mucilage showed no significant difference in the type of mucilage, however the intent to purchase was higher for breads with substitution of dry mucilage at 50°C. In relation to mayonnaise, the replacement of oil per lyophilized chia mucilage showed increased stability relative to the control sample, and texture properties similar to commercial mayonnaise. Since the addition of lyophilized chia mucilage replacing egg yolk presented negative results in texture parameters, and reducing the fat content. The mayonnaise with egg yolk replacement were better accepted than the mayonnaise with oil replacement .From this study, we verified that chia mucilage has properties that enable it to act as an emulsifier in food products. Additionally, the developed products were healthier with lower lipid content, ensuring the sensory and technological characteristics.

Este trabalho teve como objetivo analisar a operação de secagem de pasta de vegetais em leito de jorro, priorizando a manutenção das propriedades funcionais dos componentes do produto final. A pasta de vegetais foi formulada através da programação linear maximizando a atividade antioxidante. A pasta de vegetais foi obtida através de trituração e homogeneização de constituintes como cebola branca, tomate, batata inglesa, cenoura, couve e óleo de soja comercial. A viabilidade da técnica da secagem da pasta de vegetais em leito de jorro foi realizado em uma célula cônica, com 0,50 kg de partículas de polietileno, avaliando a temperatura do ar de entrada (90 e 100°C) e a vazão de alimentação da pasta (100 e 200 mL h-1). O produto final apresentou tonalidade amarelo-esverdeada, com umidade abaixo de 10% (b.u). Os teores de compostos fenólicos e a atividade antioxidante total foram afetados pela secagem ao nível de 95% de significância (p<0,05), apresentando uma faixa de (4126±79) a (5615±45 µgEAG gamostra seca-1) e de (75,8±0,1) a (83,4±0,2%), respectivamente, em relação à pasta de alimentação. A condição de operação de 100 mL h-1 a 100°C obteve menor retenção de material no leito, como também menor alteração da cor, menores perdas nos teores de compostos fenólicos e atividade antioxidante total, sendo próximos à pasta in natura. Também foi analisado o efeito da vazão de alimentação (400 e 600 mL h-1), temperatura de entrada do ar (90 e 110°C) e concentração de sólidos da pasta (5 e 10%) em leito de jorro cone-cilíndrico, avaliando propriedades do produto desidratado como compostos fenólicos, atividade antioxidante, solubilidade protéica e alteração da cor. A condição operacional com vazão de 400 mL h-1, concentração de sólidos de 10% e temperatura do ar de 110°C, foi o que apresentou valores mais adequados das respostas para o produto final tais como teor de compostos fenólicos de (6868±57µgEAG gamostra seca-1), atividade antioxidante de (61±0,2%), alteração da cor de (18±0,8%) e solubilidade protéica de (20,9±0,6%). Nesta condição obteve uma retenção de sólidos de (21,9±1,0%), e o produto final com umidade de (9,9±1,0%), tonalidade amarela-esverdeada, e reidratação de 85,4 % em relação à pasta in natura.; This study aimed to analyze the operation of vegetables paste in spouted bed drying, prioritizing maintaining the functional properties of the components of the final product. The vegetables paste was formulated by linear programming maximizing antioxidant activity. The vegetables paste was obtained through grinding and mixing of constituents such as onion white, tomato, potato, carrot, kale and soybean oil commercial. The viability of the drying technique of vegetables paste in spouted bed was evaluated in a conical cell, with 0.5kg of polyethylene particles. The inlet air temperature (90 and 100°C) and flow rate of paste (100 and 200mL h-1) were studied. The final product showed yellow-greenness color, with moisture content below of 10% (w.b). The levels of phenolics and total antioxidant activity were affected by drying conditions at 95% of level of significance (p<0.05), with a range from (4126±79) to (5615±45) µgEAG gdry sample-1, and (75.8±0.1) to (83.4±0.2)%, respectively, in relation to the vegetables paste feed. In the operation condition of 100mL h-1 at 100°C was obtained the lower retention of material in the spouted bed, as well as decreased color change, and the lower losses in the content of phenolic compounds and total antioxidant activity; these values were near of the vegetables paste in nature. Also, it was analyzed the effect of paste flow rate (400 and 600 mL h-1), inlet air temperature (90 and 110°C) and paste solids concentration (5 and 10%) in spouted bed cone-cylinder, evaluating the properties of the dehydrated product as phenolic compounds, antioxidant activity, protein solubility and change of color. The operation condition with flow rate of 400 mL h-1, solids concentration of 10% and air temperature of 110°C, showed the most suitable values of the final product such as phenolic compounds content (6868±57 µgGAE/gdry sample), antioxidant activity (61±0.2%), change of color (18±0.8%) and protein solubility (20.9±0.6%). In this condition the solid retention in bed was of 21.9±1.0% and the obtained final product with the moisture content of 9.9±1.0% and yellow-greenness color, and the rehydration of 85.4% in relation to fresh vegetables paste.

A economia da metade sul do Rio Grande do Sul é pautada principalmente na atividade agropecuária. Com uma área de 1,1 milhão de hectares dedicadas ao cultivo de arroz, a região contribui com aproximadamente 65% da demanda nacional deste cereal. Ante esse cenário, surgiu a linhagem de arroz BRS AG que foi incorporada nos programas de melhoramento genético da EMBRAPA. Devido a alta porcentagem em amido, esse grão pode tornar-se uma boa alternativa para a produção de xaropes açucarados. Sendo assim, este trabalho tem por objetivo propor um sistema enzimático de liquefação e sacarificação do amido presente na farinha de arroz do cultivar BRS-AG para a obtenção de um extrato de xarope açucarado. Os grãos foram convertidos em farinha e determinada a composição centesimal e perfil granulométrico. As enzimas, alfa-amilase fúngica e amiloglicosidase (AMG), foram caracterizadas pela atividade enzimática. Mediante microscopia eletrônica de varredura foi possível obter imagens dos grânulos do amido. Para definir a melhor condição da liquefação do amido foi adotado um planejamento fatorial tendo como variáveis independentes a relação sólido/líquido, a temperatura e a variável dependente, o teor de açúcares redutores. A etapa da sacarificação, conforme recomendação de outros autores, foi realizada em 30min a 55°C e com concentração enzimática de 4480,2U.mL-1. O diâmetro médio de Sauter foi de 0,43mm. Esta matéria-prima apresenta 8,31% de proteínas, 4,68%, de lipídeos, 77, 91% de amido, 25,20% de amilose e 52,80% de amilopectina respectivamente. O ponto de gelatinização foi atingido a 80°C em 6min. Na etapa de liquefação a maior concentração de açúcares redutores, expressos como teor de glicose (9,95µmoles.mL-1) foi obtida com uma relação sólido/líquido de 1/20, concentração enzimática de 2U.mL-1, 30min de hidrólise a 60°C. Na etapa de sacarificação, com a AMG, foi obtido um teor de 11,35µmoles.mL-1 em 30min de hidrolise a 55°C, e 0,171mL de solução enzimática (4480,2U). Houve um acréscimo de 14% na concentração de açúcares na sacarificação e 86% da conversão total do amido, nessas condições o rendimento total da hidrólise foi 4,6%. A separação cromatográfica por HPLC evidenciou a maior concentração de glicose; assim, a cultivar BRS AG apresenta um potencial importante para a obtenção de hidrolisados enzimático com alto teor de açucares redutores.; The economy of the southern half of Rio Grande do Sul is based mainly on agricultural activity. With an area of 1.1 million hectares dedicated to rice cultivation, one region contributes approximately 65% of the national demand for this cereal. Against this backdrop, the BRS AG rice strain emerged and was incorporated into EMBRAPA's genetic improvement programs. Due to the high percentage of starch, this grain can become a good alternative for the production of sugary syrups. Therefore, this work aims to propose an enzymatic system of liquefaction and saccharification of the starch present in rice flour of the cultivar BRS-AG to obtain a sugar syrup extract. The grains were converted into flour and a proximate composition and granulometric profile were provided. The enzymes, fungal -amylase and amyloglycosidase (AMG), were characterized by enzymatic activity. Through scanning electron microscopy it was possible to obtain images of the starch granules. To define the best condition for starch liquefaction, a factorial design was adopted, having the solid / liquid ratio, temperature and dependent variable, the reducing sugar content as independent variables. The saccharification step, as recommended by other authors, was carried out in 30 minutes at 55 ° C and with an enzymatic concentration of 4480.2 U.mL-1. Sauter's average diameter was 0.43 mm. This raw material has 8.31% of proteins, 4.68% of lipids, 77, 91% of starch, 25.20% of amylose and 52.80% of amylopectin respectively. The gelatinization point was reached at 80 ° C in 6 minutes. In the liquefaction stage, the highest concentration of reducing sugars, expressed as glucose content (9.95µmoles.mL-1) was obtained with a solid / liquid ratio of 1/20, enzymatic concentration of 2U.mL-1, 30min of hydrolysis at 60° C. In the saccharification step, with AMG, a content of 11.35µmoles.mL-1 was obtained in 30 minutes of hydrolysis at 55 ° C, and 0.171 ml of enzymatic solution (4480.2U). There was an increase of 14% in the concentration of sugars in saccharification and 86% of the total conversion of starch, under these conditions the total yield of hydrolysis was 4.6%. The chromatographic separation by HPLC showed the highest concentration of glucose; thus, the cultivar BRS AG has an important potential for obtaining enzymatic hydrolysates with a high content of reducing sugars.

A pena é o material queratinoso mais abundante na natureza, constituindo um subproduto de origem animal com cerca de 80-90% de proteína bruta, sendo que 85-90% dessa proteína é formada por queratina. A resistência das queratinas, somada à sua elevada produção como resíduo, vem despertando interesse na investigação de processos que possam modificar as estruturas proteicas deste material. Dentre as proteases, as queratinases são enzimas específicas, capazes de hidrolisar as ligações peptídicas da queratina, sendo produzidas por diversos micro-organismos, que se destacam, pela capacidade de utilizar materiais de baixo custo como substrato, além de possuir uma alta taxa de produção quando comparados com outras fontes de proteases. O objetivo deste trabalho foi maximizar a hidrólise enzimática da farinha de penas, utilizando a enzima proteolítica de Bacillus sp. P45 e avaliar a capacidade antioxidante do hidrolisado enzimático. A hidrólise enzimática foi avaliada através de um delineamento composto central rotacional (DCCR) 24 , no qual, os efeitos das variáveis concentração de CaCl2 (0-100 mmol/L), temperatura (35-55 °C), razão enzima/substrato (600- 6000 U/g de proteína) e concentração de proteína (10-40 g/L) foi avaliado em 8 h de reação, tendo como respostas o grau de hidrólise (GH) e a recuperação de proteína (RP). Os maiores valores de GH e RP foram obtidos quando a hidrólise foi realizada com 50 mmol/L de CaCl2, 50 °C, 6000 U/g de proteína e concentração proteica de substrato de 10 g/L, na qual o hidrolisado apresentou um GH de 8,7% e uma RP de 38,4%. Os peptídeos obtidos foram separados por massa molar através de ultrafiltração nas membranas de corte de 3 e 10 kDa e as atividades antioxidantes das diferentes frações foram avaliadas. A fração <3 kDa apresentou maior capacidade de sequestro dos radicais ABTS+ e peroxila, 90,20 e 1892,47 ?mol de Trolox/g, respectivamente. O presente estudo mostra o potencial da protease de Bacillus sp. P45 em hidrolisar a farinha de penas, visto que com o planejamento proposto foi possível a maximização e aumento de 2,48 vezes do GH e 2,37 vezes da RP, em comparação aos ensaios iniciais realizados, resultando em peptídeos com atividade antioxidante.; Feather is the most abundant keratinous material in nature. This by-product of animal origin presents around 80-90% of crude protein, of which 85-90% is keratin. The resistance of keratins and its high production as waste have been awakening interest in the investigation of processes that can modify the protein structure of this material. Keratinases are specific enzymes capable of hydrolyzing the peptide bonds of keratin. These are produced by several microorganisms, which stand out in the production of proteases due to the ability to use low-cost materials as a substrate, in addition to showing a high production rate when compared to other protease sources. The aim of this work was to maximize the enzymatic hydrolysis of feather meal through the use of the proteolytic enzyme from Bacillus sp. P45 and to evaluate the antioxidant capacity of the hydrolysate. Enzymatic hydrolysis was evaluated using a 24 central composite rotatable design (CCRD), where the effects of the variables CaCl2 concentration (0-100 mmol/L), temperature (35-55 °C), enzyme/substrate ratio (600-6000 U/g protein) and protein concentration in substrate (10-40 g/L) were evaluated in 8 h of reaction. The response variables were the degree of hydrolysis (DH) and protein recovery (PR). The highest values of DH and PR were obtained when hydrolysis was performed with 50 mmol/L of CaCl2, 50 °C, 6000 U/g-protein and protein concentration of 10 g/L, where the hydrolysate presented DH of 8.7% and PR of 38.4%. The peptides obtained were separated by molar weight through ultrafiltration using membranes of 3 and 10 kDa and the antioxidant activities of the different fractions were investigated. The fraction <3 kDa showed the highest scavenging capacity against the ABTS+ and peroxyl radicals, 90.20 and 1892.47 ?mol Trolox/g, respectively. The present study shows the potential of Bacillus sp. P45 to hydrolyze feather meal, once it was possible to maximize and increase the DH by 2.48-fold and the PR by 2.37-fold, compared to the initial tests performed, resulting in peptides with antioxidant activity.
