- ItemInfluência da composição e da fermentação nas propriedades físico-químicas e nutricionais de multimisturas(2008) Feddern, Vivian; Pinto, Stephanie Silva; Nogueira, Katiane Almeida; Furlong, Eliana Badiale; Soares, Leonor Almeida de SouzaEste trabalho teve como objetivo avaliar físico-química e nutricionalmente oito multimisturas, que diferiram quanto ao tipo de farelo (trigo ou arroz), presença ou ausência de pó de folha de mandioca e submissão ou não à fermentação em estado sólido com Saccharomyces cerevisiae durante 6 h a 30 °C. A composição proximal foi realizada segundo a AOAC (2000), o pH e a acidez segundo as normas analíticas do Instituto Adolfo Lutz (1985), os açúcares redutores por espectrofotometria, a digestibilidade protéica in vitro e a metionina disponível por método enzimático. As multimisturas com farelo de arroz apresentaram médias dos teores de lipídios (18,1%), cinzas (14,1%), fibras (7,8%), acidez (3,1%) e digestibilidade protéica in vitro (80,8%) mais elevadas do que as multimisturas com farelo de trigo, as quais, por sua vez, apresentaram teores de proteínas (14,5%), carboidratos (68,9%) e pH (7,8) maiores que as multimisturas com farelo de arroz (12,3% de proteína). A digestibilidade protéica in vitro variou de 62,1 a 81,2% e o conteúdo de metionina de 0,58 a 2,09 mg.g–1. Os açúcares redutores apresentaram um decréscimo médio de 16% em função da fermentação. A presença da folha de mandioca duplicou a acidez de duas multimisturas. A fermentação ocasionou variação máxima de 2,6 pontos percentuais para proteínas e 1,5 para cinzas, aumentando a acidez em 5 vezes, diminuindo no máximo 1,2 unidades de pH, 9,8 pontos percentuais de carboidratos e 1,26 mg.g–1 de metionina disponível, não alterando a digestibilidade protéica in vitro.
- ItemSolid-State Fermentation for the Enrichment and Extraction of Proteins and Antioxidant Compounds in Rice Bran by Rhizopus oryzae(2012) Kupski, Larine; Cipolatti, Eliane Pereira; Rocha, Meritaine da; Oliveira, Melissa dos Santos; Soares, Leonor Almeida de Souza; Furlong, Eliana BadialeThe objective of this work was to evaluate the solid-state fermentation with Rhizopus oryzae CCT 7560 of rice bran for the enrichment of proteins and the antioxidant compounds in the fermented biomass. Fermentation was performed in tray bioreactors at 30ºC for 120 h. Protein extraction was done at alkaline pH, followed by precipitation with acetone. Phenolic compounds were extracted with cold methanol. The maximum protein was recovered from after 120 h (26.6%). The content of total phenolic compounds increased during the fermentation and was maximum after 96 h, which inhibited the DPPH radical by 87%. The promising characteristics of the protein and phenolic extracts of the biomass suggested the application in the coating composition for vegetal tissues preservation.
- ItemEffect of deoxynivalenol and T-2 toxin in malt amylase activity(2010) Buffon, Jaqueline Garda; Baraj, Edlira; Furlong, Eliana BadialeThe objective of this work was to evaluate the effect of DON and T-2 toxin levels on the malt amylase activity. The malt was contaminated in accordance with central composite design experiment with DON and T-2 toxin levels until 1000 ng/g. The activities of the enzymes were evaluated by Bernfeld method. The increase in T-2 toxin concentration inhibited the a-amylase activity. However, the increase of both toxins concentration caused inverse effect. The interaction between toxins indicated synergetic effect on the b-amylase activity. An increased activity occurred when the toxins contamination levels in malt were higher (1000ng/g malt). The trichothecenes interfered with the performance of aminolitic enzymes in the stage of malting, resulting in a significant model for enzymatic activity of b-amylase.
- ItemDeoxynivalenol (DON) degradation and peroxidase enzyme activity in submerged fermentation(2011) Buffon, Jaqueline Garda; Kupski, Larine; Furlong, Eliana BadialeThis work aims to evaluate deoxynivalenol degradation by Aspergillus oryzae and Rhizopus oryzae in a submerged fermentation system and to correlate it to the activity of oxydo-reductase enzymes. The submerged medium consisted of sterile distilled water contaminated with 50 μg of DON and 4 × 106 spore.mL–1 inoculum of Aspergillus oryzae and Rhizopus oryzae species, respectively in each experiment. Sampling was performed every 24 hours for monitoring the peroxidase specific activity, and every 48 hours for determining mycotoxin levels. Results showed that the fungi species were able to decrease DON levels as the peroxidase activity increased. The 48 hours fermentation interval presented the highest peroxidase specific activity (ΔABS/minute.μg.protein–1), 800 and 198, while the highest DON degradation velocity was 10.8 and 12.4 ppb/hour, respectively in both cases for Rhizopus oryzae and Aspergillus oryzae.
